In 1647, the seigneury of Maur was granted to brothers Jean Juchereau de Maur and Noël Juchereau Deschatelets. Settlement was slow: by 1663, only three lots had been granted. A seigneurial mill was built in 1672 where Lac Saint-Augustin drains into the river, and the Catholic parish was founded in 1691.

The Chemin du Roy crossed the seigneury from 1716. In 1734, the Hospitaller Sisters of Québec became the seigneurs of Saint-Augustin. They held it until the seigneurial system was abolished, and its revenue went to the upkeep of the Hôtel-Dieu de Québec.
The current church opened in 1816, about two kilometres north of the river, on the road that would become Route 138. The village long remained agricultural. Around 1965, several religious communities built a group of some twenty white buildings southwest of the lake, designed by architect Jean-Marie Roy.
The town took on a suburban character from the 1960s, and the Les Bocages neighbourhood began to grow around 1975. Merged into Québec City on January 1, 2002, Saint-Augustin-de-Desmaures voted in a referendum on June 20, 2004, to become a separate city again. It was reconstituted on January 1, 2006.
